Fluorescein is an organic compound and dye based on the xanthene tricyclic structural motif, formally belonging to triarylmethine dyes family.
The Alexa Fluor dyes were chemically synthesized through sulfonation and additional chemical modifications made to the well known families of coumarin, rhodamine, and cyanine dyes, and to the xanthene family (of which the fluorescein, an industry-standard, is a part).
Many xanthene dyes can be prepared by condensation of derivates of phthalic anhydride with derivates of resorcinol or 3-aminophenol.
DyLight Fluors are synthesized through sulfonate addition to coumarin, xanthene (such as fluorescein and rhodamine), and cyanine dyes.
It is derived from fluorescein, but differs by the presence of four bromine atoms at positions 2, 4, 5 and 7 of the xanthene ring and four chlorine atoms in the carboxyphenyl ring.
In the reaction, the two substrates, in the presence of hydrochloric acid and acetic acid, produce a fluorone derivative that is subsequently reduced to give a xanthene derivative, which, after subsequent methylation and oxidation, leads to a xanthone with three methoxy groups.
